Thats one of the reasons spell card rules exist, so Reimu doesnt fucking die when Yuuka causes some trouble. but if a Youkai or a group of Youkai were to say "fuck the rules" and kill her, then said Youkai would probably be gang-raped by the sages. the sages are the reason the Hakurei line exists so they would probably find a replacement. im pretty sure the barrier was linked to the god of the Hakurei shrine and not Reimu, so i guess they would need to find another shrine maiden quickly before the god dies from a lack of faith.

It's the complete opposite, the spellcard rules exist to give Youkai a chance against Reimu and not just die 5 mins after coming to Gensokyo.
That's part of the canon explanation for it. The only youkai in Gensokyo that are able to come close to Reimu in actual strength are the sages, and even that's dubious.
And no, it's unlikely youkai will group up to kill the shrine maiden. Gensokyo exists as the youkai's safe haven, and going against the Hakurei Shrine is going against Gensokyo.

There's not a lot of 2hus that can kill her outside of LoLK characters, racist moonchinks, and MAYBE the sages. Marisa has already said that when serious, she's unreachable by anyone in Gensokyo, and there's a reason why many youkai and fairies are afraid of Reimu.
It's very unlikely that line in the rules refer to Reimu, and more to the generic exterminators and people like Marisa.

Not counting PC98, anyone that was currently living in Gensokyo when Grimoire of Marisa was written cannot kill her. She's completely invulnerable to even their conceptual powers. Her power is to no-sell everything.
And if you're talking about PC98, Reimu accidentally destroys the planet with the Yin Yang orb in Touhou 1's bad ending. Discounting that, Junko and co. already had encounters with people much stronger, with people that can destroy stars (Hou-yi). Whether or not a planet destroying bomb can kill her is up to debate.
