My idea then is to just copy the nemesis system. You have 50 villains each with their own tactics randomly generated. As you go on in the game they balance shifts as villains get sent to prison/escape and some get stronger while others get weaker. Then depending on the balance a villain group like the spiderman sinister 6 or a mastermand like lex luthor or joker can be generated and he becomes the main rival.

I don't know shit about Power Girl so the first thing I'd do is call up Kevin Smith and scour forums and the internet for committed virgins who love comics to tell me all about the character. Tell me all about her flaws, her strengths, her attitudes and aptitudes, her personality, her hobbies, her best assets, her closest relations, her worst enemies, ect. I'd capitalize the marketing on how strong wymyn are and how you play as a badass bitch!!!1! Make the actual game a story of growth where Power Girl fails to save civilians and they die in front of her. She starts to suffer from survivor's guilt and it really fucks with her and as she tries to violently lash out at bad guys she realizes that she's kind of a bad guy too. She works through that shit mission after mission and comes to peace with the fact that she's kind of a mercenary who kills people to keep a larger number of people safe. The people she saves may be as shitty as the bad guys, but it's all for the greater good. Then her puppy gets stolen and the game ends and the sequel talks begin...
