Like others suggested, a story oriented game with focus on witty jokes, maybe a point and click adventure with quirky humor. It's of extreme importance that it retains realistic looks for the characters, that means old ass adults dressed as children and the same actors being used for different characters, it adds to the charm.

what about making the bald guy and el quajinais the main antagonists, making the bald guy plot that he wants la vecindad again, and gets el quajinais to work for him and his goons to get it, but while that happens, el quajinais is doing his own evil doings, and betrays him at the end of the game, and becomes some kind of final boss quajinais

you should consider the mailman being an occasional optional fight whenever he appears, if you defeat it, he gives you important items. i never played any of the mother games so i don't know about their stats or if they have armor items to increase attack or defense
adding goons in la vecindad sounds great, is a reminder for the player to know the plot is advancing adding more difficulty or amount of them to fight depending how far you go in the story, a good way to boost levels, maybe give them some simple strategies that quajinais gonna use in their final battle so players can get some tips in how to deal with his moves

you can make el sr barriga be the middleman first as a midpoint of the plot only to discover that the true antagonist are either you mentioned, after defeating el sr barriga, ñoño joins your team as a way to pay for his father's mistake even though sr barriga was not really a bad person, hew was just doing his job

a persona style game where you play as el chavo teaming up with quico, chilindrina and the other kids to save the neighborhood. the first part of the game is to help don ramon to pay his rent by gathering money to help him. of course there is a day system that also counts as a time limit how long you can help don ramon in paying his rent, failing on this quest don ramon is forced to move out taking away chilindrina who is part of your party.
oh, i don't remember much about the show, sorry for that. you can work this around by first making everyone to believe sr barriga is the enemy because he is forcing don ramon to move away for not paying his rent and by defeating him you open the path for calvo to buy it since barriga is incapacitated to do his job.

sounds better than what i had in mind. you can even make don ramon hide in the barrel sometimes, adding some dialogue when you go to the barrel when trying to get items saved in it, of course if you plan to make the barrel be the chest to keep all your items gained from battles that is. he could be like the store owner but he doesn't charge you to withdraw or deposit items, just a fun feature, he could even give you hints or even give you items he found . i dont know much about the programming so if this all sounds complicated, be free to omit them, i really like what you are doing.

how many you planning to add to be in your party besides chavo and quico? you should make quico have his "squared" ball be his weapon of choice.

AFAIK, there isn't. There is for a few Chapulin Colorado episodes. I don't think is even know by the general people there, but, the series is like religion to Brazilians, and since Brazil has the biggest colony of Japaneses outside Japan, and Brazilians are everywhere in Japan too, it's not uncommon to some Japanese be familiar with it.
