Officially it's short for Dual Screen. But at Nintendo it was unofficially short for Developer's System, since it was easy to develop games for, and the touchscreen and microphone encouraged all kinds of new game mechanics.

I have one that looks like that and it still works.

The 3DS actually does have three screens though. The 3D effect is achieved by having two screens superimposed on top of each other with slightly different viewing angles.
