Safe Mode only brings the core essential components of Windows online from a protected state. It does a rudimentary check to verify integrity to core system components to prevent mimicking.
You do not want to do scans while in normal mode because many pieces of malware are specifically made to hide itself from scanners. Starting in Safe mode should be step #1 to any scan you do because that prevents anything that has injected itself into your system startup list from starting.
It doesn't make it more likely to detect it, but it makes it less likely that it will prevent you from detecting it.

You can with more than one antivirus/malware program because the definitions aren't shared between them. The database for one isn't the same as the other, and they will detect different things.

No, Sandboxie simply limits which sections of memory a process and all child processes can use. It uses its own sandbox-versions of windows daemons to handle processes that use that section of memory, so the process itself should think that it's actually running on native Windows itself.

This doesn't work with everything, and you'll find tons of programs that will flat-out refuse to work because the sandbox doesn't give them enough flexibility to operate, but Games and Installers shouldn't be one of those things. Unless you have a game that has absurd requirements or requires special windows components, then it should run as if it's running natively.
