I think Dante knew and that V even told him when he walked into the shop. Him not knowing at all until the end would make Dante and Trish the most amoeba brained retards imaginable, plus it works better for character building if the reason Dante's so pissed at Vergil at the end is because he was willing to give V a chance but he just ends up merging back anyway.
I think the "Was this your plan all along, V?" line confirms this.

Yeah, everything points to Dante knowing from the start besides a couple of minor points that could easily be explained, like Dante saying he "realized something was off" to V's summons (he might just not have realized the summons were Nelo Angelo's thoughts) and him laughing off V's attempt to stab him through the head with Sparda (it would be incredibly dumb to brush that off when Dante's still not sure how much of Vergil is in V but then again the brothers skewering each other might as well be a greeting).
I think the most obvious evidence is Trish telling Dante "V told me everything" after the first SDT fight and Dante doesn't question it

At first, sure, but power corrupts. Even if he didn't "lead" the demons, he would still rule them just by virtue of being the strongest. They would constantly try to overthrow him in the power struggle, not to mention Mundus wasn't the only one who wanted revenge against the Sparda bloodline. It actually started with a scene that seemed to show Dante getting killed before a weird guy entered his shop, created a bunch of tension, flipped his desk, then a bunch of demons suddenly impaled him. THEN Dante shrugged it off and had his action scene.
That's the point. You contrast the dark mood with Dante's brash attitude plus the weirder elements of the series.

DMC3 begins with a creepy dude talking to Dante and DMC1 starts with a crazy woman throwing a motorcycle at Dante and trying to kill who him, who he agrees to help afterwards, and Dante doesnt swing in a stripper pole in cutscene that's just gameplay that he does with any pole in the game, and when you aren't fighting crazy demon characters, you have this dead city with blood all around, and a tower that demon worshippers build, and DMC3 was gloomy for most of the play time while in cutscenes and bosses it felt more lighthearted

It took Dante ten extra years of training and hunting demons before he could confront Mundus with the full power of Sparda, and even then he ultimately needed to seal Mundus with Trish' help. Vergil would have lacked those ten years and wouldn't have had any way to seal Mundus.
