The game "resets" every time you beat him so he's always available. To get all endings though you need to not have the Void Heart and beat him. If you have it tough shit, you need to make another save.

Yep. As long as you don't have the void heart before fighting him, you can get all 3 base endings. After you get the first one, you can simply go pick up the item and beat him again.

A prequel is, by definition, something that came out after a game that it is the backstory to. There is no "in the release order sense". Whether you call something a sequel or a prequel, it both means that it came out after something else.
