The centipedes are yokai featured in Buddhist beliefs representing stagnation, a running theme of the game if what another user said is right.
Kuro is the divine child because the dragons bloodline was passed to him sometime after Takeru died, the rice girl is the result of the monk's attempts to create their own version of immortality. She's the only one who survived the process, all the stone Buddha statues you see on the way up are each for a dead child and she talks with their ghosts in the halls of illusion, as does Kotaru.
Mist nobles are people corrupted by the fountainhead waters, similar to the people in Mibu village. Why they turned the way they did isn't really explained.
Shichimen warriors are lingering Ashina warrior spirits if I recall correctly.
They're samurai belonging to the ministry of the interior that is warring against Ashina, they're who the Tengu refers to as rats. Most of the short dudes also work for the ministry.
Genichiro could use lightning because the game has supernatural elements and he trained with Tomoe, who had mastered the art of wielding lightning.

Pure speculation, but the reason the Divine Dragon may have been missing his arm is because his severed arm brought down from the heavens could have been the Everblossom sakura tree talked about in the plot.

(Speculation) During her travels she ended up meeting the jar noble which in exchange for where to learn more techniques she taught him her personal combat art, "Flowing Passage". I believe she ended up at the divine realm as that is the most common area for lightning based attack, plus the jar noble who knows Tomoe's personal sword style also knows of he divine realms location and existence

You need a centipede to be "immortal." The dragon water just gives unnaturally long life and turns people into fish. The red nobles are the oldest and furthest gone. They're devouring the younger nobles because they desperately crave youth and immortality.
The mist nobles' goal is to become immortal by becoming carp. They need fish scales to do this, which is why they are eating other warriors.
Sekiro's immortality is very different from centipede immortality. Sekiro is capable of death and rebirth, but the nobles are only undying. Wounding them outright will still kill them.
