Proper spore 2? Planet size needs to be hundred or even thousandified, this is just bizarre. Maybe warcraft 3 like gameplay? allow building everywhere, and don't use FUCKING FOOD SPICE as resource. Use something that tech couldn't produce. exotic matters or so? Most important, don't let EA publish it, they want amends and their amends mean to casualize it into oblivion. That's just my brainstorm though.

to be entirely fair, tech couldn't reproduce spice in the Dune novels either (at first)

Dune-verse spice is an extremely complex organic molecule that enables various super-human feats when ingested, including the future sight needed to properly operate FTL ships

Depends on what you mean by "spice". If we're talking about food spices like on Earth, then yeah, those are plants. The spice in Spore was pretty clearly something that had to be mined and I don't think they ever really got into what exactly it does or why it's valuable. Most likely they just needed a name for the currency and they decided on giving a nod to Dune. In Dune, spice is the chemical that allows for FTL travel because it turns people into weird psychic alien people who are super smart or something.

the super-smart thing is a whole other part of the setting involving 10,000 years of eugenics turning a select part of the population into human computers, as actual computers are illegal
there's no aliens in Dune, just the navigators who, after ungodly amounts of spice-exposure combined with constant 0g environments turn into mutant fish-men
yeah the only spice that's mined would be salt. Couldn't they just used Actinides or something? Uranium, Plutonium, Neptunium, Curium... yknow.
Yeah the problem is if you can clone beings or even 3D print them from scratch, why can't you 3D build the molecule or, lower tech, put the genes that tell the cell to make the producing enzyme in a fast grower such as E. Coli, from there you can use brewery-like production.

Eventually they do manage to synthesize the spice but during the first few books there are a few massive hurdles against it
1: Computers are illegal in the Dune universe, as in flagrant disregard gets your planet bombed from orbit, as such actual synthesizing molecules is a lot harder
2: spice is incredibly complicated and supernatural in origin, there's hints that the sandworms that produce them are also ancient lost biotech
3: control over spice is the key to controlling the galaxy, only the people who control Arrakis (the planet where it is produced) can allow for experimentation (as trying to breed sandworms off-planet invariably fails) and they have exactly zero reasons to do so

this would require experimenting on a mile long creature with free roam over an entire planet-wide desert that fucks with technology something fierce, while dodging religious fanatical natives who'd kill you for even suggesting it and are the only people who can actually survive in the desert, go against the direct decree of the Emperor of all of mankind AND somehow manage to get there, with your equipment while the people who hold an absolute monopoly on all FTL travel would be legally obligated to toss you out of the airlock if they found out what you're trying to pull
they actually try in universe but there simply is no way to get the proper samples

it's only found out during the course of the first book that the spice does not originate form the sandworms but rather from their microscopic larval forms so even managing to capture a worm does nothing
could be an interesting story for a vidya, where you have to collect tissue from such worm somehow? I'd suggets shooting it to bloody viscera because mandatory bossfight.
the worms have skin tough enough to withstand anything short of nuclear weapons, and using a nuke for any reason gets you MAD'd by all noble families in the galaxy per thousand-year old treaty
Dune-universe is actually well thought out, so most means of getting worm tissue is already explored in universe at one point
How about making a giant mechanical worm to stealth by and when a male wants to jump the mecha, a hole opens to chop the soft penis off. Hey, it's not stupid if it may work.
