1. Centipedes
2. Mibu/fountainhead water (and centipedes lay eggs in this. Also prolonged exposure + Water of the Palace turns you into a fish dude)
3. Divine Heir contract

I mean I still beat it without a hold up. But the game isn’t nearly as bullshit as previous From games were about hp bloat and 1-shots. Almost being 1-shot is the norm in Sekiro. Makes additional play throughs fun.

Drinking the water makes you crazy and slowly turn into a fish like those at the fountainhead, but it also makes you somewhat immortal. It's like a poor man's dragon blood. The monks tried to make REAL dragon blood tier immortals with it but most of the kids like that died (including Robert) except the one you meet in the game. The people of Mibu were pseudo immortal (and also completely insane) because of the water but not totally immortal like the Infested.
The infested are a different kind of immortal being that comes from hosting the centipedes that hatch from the eggs in the immortal waters. It's a bit more effective, you can get your head cut off and still keep going, but being infested with a giant immortal centipede puts you in constant excruciating pain that you have to live with FOREVER.
