anyone can be defeated with 1 quick suckerpunch from something they didn't unexpect, but Dio is still a vampire. His weakness is the sun, and be quick enough to decapitate him before he stops time (and he can't heal fast enough when time is stopped hopefully) and just push him into sunlight lol.
If there only was some creature or person who didn't have blood but had UV powers or "powers of the sun" and was really quick.
Oh I know, The Flash can, before DIO stops time, put DIO at a spot of the earth where there is sunlight and BAM! dead lol

literally any trained sniper could kill him. Star platinum destroyed Dio's skull and he lost control of his body, including the time stop ability. .50 BMG would have the same effect. Just neutralize him and put him in a cage until sunlight

I assume since the world is essentially the same as star platinum, it would be able to catch/dodge the bullet like jotaro did with the pistol in episode 1.

Definitely Diavolo. Though Dio can stop time and all that, Diavolo has a much more functionally useful stand that can hypothetically nullify all of Dio's movements and leave him perplexed. What's worse, timestop is actually a hindrance as the moment time is stopped would be skipped, frustrating Dio even more.
