She had already been labeled Moon Princess as was guaranteed to be Naruto's love interest during the first draft, when Sakura and Sasuke didn't even exist yet. In japan "heroine" can mean that but can also mean the (romantic) companion of the hero. Hence why Orihime and Hinata are the "heroine" of their respective series (If you don't believe me, just go check for the interviews of their author).

I think he meant that he planned the main plot but the subplots with relatively minor parts are managed according to however he wants it to. As an example,he didn't bother to expand on Sakura's backstory as she was really unpopular,if she was he would have.
