An isekai where the main character absolutely hates the fact he's been isekai'd, not because he had a particularly good life but more because he see's being isekai'd not as a second chance but more like the universe looking down at him, hurting his pride, and the whole story is about him doing his best to get back to his normal life full of ups and downs and his rejection of the happiness and comfort the world he's been isekai'd to brings.

A boy falls in love with a girl.
Unable to confess, he is gifted with by a deus ex machina with the girl's phone number. Never minding the strange area code, he immediately calls her, and is overjoyed to find out that she has a crush on him as well.
But, the next day, when he goes to recount the previous day's confessions to the girl, he is struck by a truck and killed. After some investigation, he finds out that he died and is now longer in the same city he was when he died. In fact, he isn't in his old universe at all. He soon finds the girl's alternate universe counterpart, who has fallen in love with the MC's own AU self, who too is blissfully unaware of her crush.
Hijinks ensue as the two strike up a deal to find a way home for the MC and to give each other their darkest, most private secrets in order to equip the other with the weapons they need to conquer the heart of their other selves. While the two chase their respective goals, DRAMA ensues as they begin to fall in love with each other instead and question the NATURE of LOVE.

His foster father explains that humans have long since been extinct from their world, though he doesn't know why or how exactly the MC came to them. Faced with an existential crisis, the MC leaves the village the first time in his life to try to understand the world, the role humans once played in it, and how his existence can change it's whole foundation.
A Japanese is isekai'd to... FRANCE! He gets teleported to the guy from poptepipic's house. The whole anime is in French with no subtitles except for MC who must conquer this new and foreign environment, eventually aiming to land a job at the local bakery and marry the baker's daughter Areesha

A self-aware comedy isekai where someone thinks they were transported into an MMO, but instead they were transported into a hentai game and gender swapped. Think some poor salaryman Japanese fuck is transported into the world of Rance as a beautiful woman (but still has overleveled broken strength) and then has everyone in the game constantly lusting after him, and he hates it.
I wish they would steal some of the Isekai ideas from China/Korea.

It could either be a comedy anime or get really dark. However, why not a little of both? The dark undertones of a young demure Japanese divorcee having created a big barbarian woman in an MMO to cope with her own inadequacies has some depressing shit there by itself . Not only that but Kanna could also lose it when she finds out her strength makes her a potentially brutal opponent and rage the fuck out. But I like the idea of the anime being more on the wholesome and funny side instead. Like, JRPG's and pretty strict regarding class equipment, so what if Kanna can't use anything other than skimpy barbarian leather armor?, anything else that's protective she tries to wear (because she's shy) just bursts into flames.
With that kind of premise I'd expect an ending where she goes back to her original world after gaining confidence in herself, but that also depends on what tone the series is going for.
A post isekai where a guy wreaks havoc in the real world after coming back from the isekai world as a demon lord with a large army.

Going wholesome it could go either way depending on if she actually finds legit friends in the Isekai. Depending how her personality evolves she could find a way out taking a learned barbarian brashness with her like you say. Or she stays because she finds out she likes being a muscle-girl in a loincloth killing shit for coin and glory. If you wanted to go shades of dark, her douche-bag ex could also have been transported into the MMO as a plot twist being either someone relishing it or being terrified like she would be in the beginning. Depending on were you'd want to take the message of the show regarding revenge, a depiction of that Gaman thing Japanese people take to, or simply moving past personal bullshit and such, is she and her ex start knowingly or unknowingly working together in some way.
A post isekai where the protagonist comes back with the stats and skills they had in the game (excluding magic).
Isekai about a mecha MMO where the guy eventually comes back and tries to build his own real one with the engineering knowledge they learned in the years they spent in the game world. Would lead to an explosive ending.
