Because Diavolo is outrageously paranoid. The man believed that Trish was a potential weakness that someone could use to both find him and uncover the secret of King Crimson, and then his eternal life on the climax would be challenged. By eliminating Trish, that chink in his armor would be filled.

The problem was never Jolyne. Even the people who shit on Part 6 usually say it was in disservice to how good Jolyne was.

It was just a bunch of unsatisfying fights, a very boring setting, and a weak supporting cast. Every other part has some positives in spite of its flaws and SO only has Jolyne. She can't carry such a mess, especially Pucci.

A big problem with Pucci is Part 5 has the most kind of unearned feeling asspulls and Pucci is just asspull incarnate. When most the readers went straight from 5 to 6 it got tiring.

He didn't know what Walking Heart could do, and purposefully set up a trap so when Hato entered the doorway she'd melt, and Damo would kill her to make Norisuke fall into despair as punishment for getting involved and helping Josefumi to kill so many of his associates. Gappy got around this by sending her inside in a bubble, then when she escaped again and Damo sealed the exit, he planted another trap by setting Norisuke on fire and throwing him in the fireplace. When Hato on the roof approached the chimney, Vitamin C will attack from the chimney and he'd win. Hato won by listening to Josuke's soap bubbles popping under the roof above Damo, so Hato just stabbed where she heard the popping, lancing Damo through the head and deactivating Vitamin C. Then Gappy chased him down and finished him off.

Giorno vaccinating himself with the snake's blood is the closest thing to a asspull since, as far as I found in my 30 minutes of research I couldn't find any place that claimed any vaccines like this are in use. Let's not even mention how incompatible the blood of a snake would be to the blood of a human. So unless you want to argue that Giorno took out the dead proteins of the virus from inside of the snake's blood and then injected himself with them it wouldn't work, and even then he wouldn't become immune since he was already infected by it and one dose of proteins is not enough to gain immunity.
So yeah, you were only half wrong, half of the snake brick thing is an asspull but the other is just Araki loving his quirky facts.

What Araki was referencing there is the process by which antivenom is made. Small traces of venom from a snake is injected into a host domestic animal and blood is collected from the host animal and the venom's antibodies are collected and purified, and are used in extreme cases to treat dangerous snakebites and stings and such because of the danger.

So in Giorno's case he created a snake inside the area that was already infected with the virus, and as such the snake developed antibodies to combat the virus naturally. When the snake bit Giorno, these antibodies were injected into him through the snake's venom, which he likely engineered to be harmless to humans. As such Giorno would receive antibodies for fighting off the virus, created from the virus itself by a host animal.
