She's old, and she worries about her fading looks. It's a play on the classic "Does my ass look fat in this?" question that women will often ask.
The joke is magnified by the fact that this women is the Elastigirl, who a few scenes earlier transformed herself into a boat. She possesses complete control over her body, yet even her body is affected by time. It's a humanising moment where the viewer is reminded of how powerful vanity is, that it can even defeat a super-human.
This is the essence of "The Incedibles". We are introduced to a family of super-human, who have been forced to hide their identity. Confronted with pride, vanity and the need to fit into the norms of society, they start to lose their identity and their family life becomes miserable. It is only once they embrace who they are, that they pull through. In contrast, Syndrome, the villain of the story, has failed to embrace his identity, and tries to become something he is not. This is what in the end kills him, the cape, a symbol of his pretentiousness.
