Honestly, I think George wrote that quote with the intent on disproving it later on. We're led to believe that Targaryens are some extremely willful people whose natures are absolutely unalterable. In reality, there may be a genetic predisposition to mental illness but the mental state of the Targaryens, like anyone else, is largely dependent on circumstances of upbringing, traumatic events, feelings of worth, etc.
The state of Targaryen "madness" is the result of things like rampant sycophantry, extreme haughtiness caused by their station in life and the fact that they're socialized to believe themselves as the children of destiny with Blood and Fire being the tools of their becoming.
Take a look at some of the later, great Targaryen monarchs: Daeron II, Aegon V, Jaehaerys II. These aren't the same heroic, willful warrior-kings that greatness implies. These are humble, self-aware and changeable men who benefited from considerate upbringings and the company of thoughtful people. I'm sure all three had their flaws, holes in their personality that made they're lives more difficult as well. They weren't born entirely good.
Now look at the description of The Mad King. There we no hints of his insanity in his youth. He was actually likeable, charismatic and generous. He was also arrogant, extremely sensitive to the constant, unwarranted praise given to him by his followers and increasingly ignorant of his councilors' concerns. This sounds a lot like the characterization of Daenerys.
And what of Rhaegar? He was described as being a great person by all accounts but he also believed so much in his prophetic destiny that he blown an already tense political situation into a destructive war, and when the time came, his destiny was ended cut short by Robert's warhammer. This was a man who cannot at all be described as entirely noble or unhinged by an honest judge.

Yes, there are 2^100 equiprobable outcomes.
But the event of "getting exactly two heads" is a bunch of outcomes. The heads could be on the 2nd and 51st try, or the 1st and the 100th, etc. That's 100 choose 2 = 100*99/2 = 4950 to get it.
On the other hand, there is only one way to get 100 heads in a row. Same for 0 heads in 100 tries.

It was too late at that point, he broke beyond repair the moment he sold his mother's crown. Rhaegar is an example of how expectations can cause a person to go insane. He grew up thinking he was the fulfillment of the Price that was Promised prophecy which was something that was first brought up to his great-grand Targaryen. He came to see his children as the fulfillment. Daenerys ended up with that burden. Targaryen mental stability was already being questioned at this point but the ominous messiah prophecy was the final nail in the coffin for the sanity of their house.
