Audience expects to laugh at Late Night talk shows, audience already primed for laughter from jokes earlier in the show AND earlier in that very segment, the live video call-in things makes it feel like a bit because people are expecting a punchline, Jerry usually only does comedy, hard to separate Michael from the character of Kramer, Michael doesn't actually seem sincere, and there's probably some nervous laughter from the ridiculous nature of the situation.

When you're with a live studio audience, you aren't thinking much about what is happening - you just go with the flow. The audience was just following along with the show. It's entirely the fault of the production side of things.

The awkward pauses due to the video-delay don't help, because the audience naturally wants to fill pauses with laughter. Eventually they figure out there's no funny angle.
