That being said, I would say Punisher could take Snake on. Frank could hang in a balls to the wall fire fight, they are both highly trained soldiers so Frank can hand with Snakes CQC. Really, the villains in the Marvel universe most likely gives Frank the edge over Snake since he deals with that shit all day.

Snake is incredibly accomplished and has literally superhuman strength and durability on par with Cap. Frank has more hands-on experience with people his superior, though. Honestly? Experience isn't enough of a factor in this fight to give Castle the edge. Snake beat two different Big Bosses, after all, each of whom pretty much qualifies as a Castle in their own right. Snake's superhuman qualities just help him too much.

Frank is a guy who only wins via authorial fiat and is an 80s character woefully out of time. Snake is constantly active soldier who fights real enemies not petty drug-dealers or whichever barely trained mafia goons Frank has for stationary targets this month.

I dunno. Frank has been able to hand with the likes of Daredevil and Bullseye. Snakes ninja flips and marksmenship is not something he would be unfamiliar with.
Definitely going Snake with this one OP. Most of the time Frank is taken on mobsters and street thug types. At least half the time his opponents don't have a chance, his very skillful proficiency with firearms and combat logistics towers over those he targets most of the time. These are the two main edges he relies on to do his job. It's pretty much shooting fish in a barrel. That's not to say he doesn't have more to his skillset, but those two are Franks meat and potatoes. Were as Snake is an elite operative; a master of stealth and CQC. With an exaggerated ability to detect enemy combatants that aren't stealth ninjas like him. Even with putting Twin Snakes aside () I'd say it's not much of a competition. Snake can take someone on like Gray Fox and survive. The MGS in world, regards to Snakes abilities, is much more exaggerated than 616 and ennis-verse in the regards of Frank's abilities mostly. Of course their is stuff like that Punisher animated film (Avengers Confidential: Black Widow & Punisher), which is also pretty exaggerated ( Which in itself may not make be as justified in Frank's case, Frank was just in 'nam as a Marine. He was naturally gifted and skilled at war, but still didn't not on the level of highly trained elite-super-soldier-ninja-operative best of the world kind like Snake.) But generally It's a non question for Snake to be able to take Frank unawares and disarm or kill him some how, once he got close or was in a vantage point. And of course Snake could take on Frank in an unarmed CQC fight between the two of them. Now of course this next point in't very fair because MGS is a videogame, but that's just how it is: I don't see Frank ever taking on a Metal Gear directly like Snake has. If Frank had to deal with one in his own story, he would do it in some way that allows for his power level, some non-direct way to cheese the battle, that's how he defeats a lot of the opponents that he is obviously outmatched by.
Hard to say. Snake is more skilled and probably a little smarter but Frank is basically magic depending on who is writing him and is able to stand up to the likes of Doom, Ghost Rider and everyone else he should logically stand no chance against. I would go with Snake for most scenarios.

Snakes lives in a world where mechs, nanomachines and advanced powered armor exists that he has access to, which would him the advantage. Granted, the Punisher lives in a similar universe in terms of technology, but he mostly uses conventional weapons unless he has Chip or other characters offering to sell futuristic tech. Given then, he barely uses them due to cost, maintenance, or because it's too difficult to use in most situations.

Realistically Frank & Snake are just as tough as each other in terms of endurance and grit, I think Frank would have a small advantage in raw power but Frank time and time again gets his ass handed to him by enemies with superior technique in a straight fight and Snake has undeniably the greater technique, CQC being supposedly the ultimate thing in physical combat.
I think it would be a close one and would end in a shirtless fistfight after they shoot, stab, burn, bludgeon, frag, bomb and blast each other to hell.
