Given how easily MacFarlane does the Peter voice and has kept it consistent, I think it just comes down to having the right mouth shape and voice, rather than technique.
I'd say the hardest voices are the ones that are basically the natural voice of someone who sounds really distinct, like Billy West as Fry.
