I would imagine that over the years there would be different incarnations of the Spectacular Six, but they're usually always led by Octavius due to his sense of duty to stopping Web Shooter from hurting another person. You could always work in Sandman, Electro, Vulture, Chameleon, etc. into the different rosters.

You'd probably have to reverse the Li and Negative personas. Have him be a slave on a human trafficking ship who kills one of the crew members, taking his identity and working his way up in local gangs in New York. Eventually, in order to prove his loyalty, he gets experimented on. This gives him a split personality, with this new "Mr. Negative" personality seeking a way to help people rather than hurt them.
You'd spin the Li/Negative personalities on their head, which could translate to E3 rather easily.
