The only American animated dramas I can think of are American Pop by Ralph Bakshi and Sealab 2020 by Hanna-Barbera.

American Pop was good, but done entirely in rotoscope; it might as well have just been live-action since it was already filmed entirely in live-action, anyway. The animation aspect adds little to it and isn't utilized for any meaningful effect.

Sealab 2020 is notoriously boring as Hell and is only remembered at all because it's parody, Sealab 2021, was good... and it was a typical edgy animated comedy series like every other American cartoon made for adults.

What do you want from a cartoon? Dead-pan serious drama? Nihilistic thrillers? You want Mickey Mouse to deal with Minnie's stillbirth as they both decline in sanity and climax by killing Donald in a satanic sacrifice orgy? It's just not in the nature of cartoons to deal with intensely serious subjects without it becoming absurd or disturbing to the point of extreme niche. A majority of people want to feel positive emotions when watching cartoons, it's only when cartoonists make cartoons for other cartoonists or snobs that more "real" subjects are explored with more depth. If you want serious subjects dealt with in a cartoon, you do it under a veil of comedy or satire so people can consciously brush it away as jokes but unconsciously accept the message or truth. I think cartoons are still too new and fresh in the collective unconscious to become a medium which is truly universal and can deliver any message in any tone. As long as there's still people whose kneejerk reactions to cartoons are "haha funny and dumb", you can never get past uncanny valley, in this case being visually unrealistic characters doing realistic things, and for that reason cartoons will always have to have, at the very least, moments of comic relief or some kind of redeeming charm.
